应用ISSR-PCR对西伯利亚红松19个种源的遗传多样性分析

摘    要:应用简单重复序列区间扩增多态性(inter-simple sequence repeats,ISSR)技术,以引种俄罗斯的西伯利亚红松(Pinus sibirica Du Tour)不同地理分布区的19个种源为材抖,筛选出12个引物,获得总位点数148条,其中多态位点数136条,多态位点比率为91.89%。遗传多样性研究结果表明:种源的平均多态位点比率为26.48%,具有高的遗传多样性(Shannon指数(Ⅰ)平均值为0.1563);种源间存在一定程度的基因流动(Nm为0.2461)和遗传分化(Nei指数平均值为0.1068,Gst平均值为0.3298);种群内的基因多样性占总群体的67.02%,种群间占32.98%。综合UPGMA聚类分析、地理变异规律及生态类型,种源划分结果为:西萨彦-阿尔泰山地生态区种源;东西伯利亚南部高原生态区种源;西西伯利亚平原生态区种源。

Abstract:Twelve primers were screened out with ISSR technique, and 148 sites were obtained totally from nineteen populations of Pinus sibirica Du Tour, an introduction as material, in different geographical distribution zone. 91.89% of which, namely 136 sites, belonged to polymorphic sites. Result of genetic diversity suggested that the ratio of polymorphic sites of populations was 26.48% in average, and the genetic diversity was abundant judging from the average value of Shannon index 0.1563. Gene flowing and genetic differentiation existed among populations considering from the value of Nm was 0.2461, the average value of Nei index 0.1068 and Gst index 0.3298. Genetic diversity among populations possessed 67.02% of the total and genetic diversity between populations possessed 32.98%. The whole populations were divided into three ecological regions according to the result of UPGMA cluster analysis and the rule of geographical variation and ecotype, which were populations of ecological region in Xisayan-Altai Mountain, populations of ecological region in southeastern Siberian plateau and populations of ecological region in west Siberian plain.
