胡杨、柽柳总RNA提取方法的建立

摘    要:以胡杨和柽柳的叶片为材料,建立了一种从植物组织中快速分离总RNA的方法。该方法具有三个显著的特点:RNA提取周期短,应用改进的LiCl沉淀RNA方法,仅需l0min即可;方法简单易行,仅需要几种常用试剂,操作步骤简单;提取的RNA杂质少,得率高,并能在提取过程中较有效抑制RNA的褐化效应。RNA质量可以满足cDNA文库构建、DDRT—PCR等要求。

Abstract:A method of rapid isolation of total RNA was developed by using fresh leaves of Tamarix and Populus euphratica Oliv as test materials. This method has three obvious advantages: the cycle of isolation is short, if using improved method of RNA deposition by LiCl it only need 10 minute to deposit RNA; it is simple and only need some kinds of common reagents; isolated RNA has high purity and browning effect can be inhibited by this method. The qualities of RNA can meet the demands of cDNA library construction and DDRT-PCR reaction.
