摘    要:为了解决利用普通石蜡切片观察植物样品需要进行脱水、浸透、包埋操作且耗时长的问题,以高山红景天为试材,对其根和茎进行冰冻切片,过程简单有效且图版清晰,在1d内即可获得高质量图片。冰冻切片的关键步骤是选择合适的甘油浓度、切片时采用的温度及展片技术。

Abstract:It needs dehydrating, infiltrating and embedding before observing when observing plant specimens with paraffin sections. Aiming to solve this time consuming process, cryo-sectioning technique was used to observe stem and root structure of two-year-old Rhodiola sachalinensis A. Bor. The operating process is simple and efficient, and clear plates can be acquired within one day. The key step is to select a proper concentration of glycerol, a suitable temperature of freezing and an appropriate way of section-flatting.
