高山红景天叶片愈伤组织诱导与植株再生

以高山红景天叶片作为外植体,研究了影响愈伤组织诱导、芽增殖、生根的主要因素。结果表明:高山红景天叶片切块诱导愈伤组织的最佳培养基为MS 6-BA 2mg/L NAA0.5mg/L,诱导率达88.33%;高山红景天叶片切块经诱导可产生3种色泽的愈伤组织,其中只有绿色愈伤组织诱芽容易,绿色愈伤组织在附加1mg/L6-BA 0.1mg/LNAA的MS培养基中有利于芽的分化和增殖,培养7周左右,平均每个外植体分化形成的再生小植株数达到21.33个;增殖苗转入1/2MS培养基中生根率高达100%。

Callus Induction and Plantlet Regeneration from Leaf Explants of Rhodiola sachalinensis A. Bot.

The main factors affecting callus induction,bud proliferation and rooting were studied using leaves of Rhodiola sachalinensis as explants.Result showed that the optimal medium for callus inducement from leaf segments was MS supplemented with 2 mg/L 6-BA and 0.5 mg/L NAA,and the callus inducement rate reached 88.33 percent.Yellow,green and red callus could be induced from leaf segments,but only green callus could be induced to bud easily.The MS medium supplemented with 1 mg/L 6-BA and 0.1 mg/L NAA was favorable for the differentiation and proliferation of buds,and the average regeneration plantlets for each explant was 21.33 after 7 weeks of culture.All the regenerative plantlets could take root after being transferred to 1/2 MS.
