摘    要:为获得优质的愈伤组织,以青钱柳嫩茎和叶片作为外植体诱导愈伤组织,研究了不同的消毒方法、基本培养基和植物生长调节剂对青钱柳叶片和茎段愈伤组织诱导的影响.结果表明,以青钱柳茎段为外植体诱导愈伤组织,最佳的消毒组合为70%酒精10 s+0.1%升汞4min,污染和死亡率最低(18.0%),诱导率最高(95.0%).以青钱柳叶...

Abstract:Stems and leaves of Cyclocarya paliuru were used as explants to induce callus in order to obtain better callus.Effects of different disinfection methods,basic culture media,and plant growth regulators on callus induction of C.paliuru were studied.When stems of C.paliurus were used as explants,the best disinfection combination was to treat the stems with 70% alcohol for 10 seconds and 0.1% mercuric chloride for 4 minutes,with the lowest contamination rate and mortality of 18.0% and the highest germination rate of 95.0%.When leaves of C.paliurus were used as explants,the best disinfection formulation was to treat the leaves with 70% alcohol for a very short time and 0.1% mercuric chloride for 4 minutes,with the lowest contamination rate and mortality of 13.7% and the highest germination rate of 96.3%.The optimal media for callus induction of C.paliuru using stems and leaves as explants were modified MS and MS respectively,in which the highest induction rates were 95.56% and 96.37% respectively.The best combination of plant growth regulators was 0.5mg·L-1 KT plus 2.0mg·L-1 2,4-D,and the induction rate reached 97.5%.
