Abstract:An experiment was conducted to establish a RP-HPLC method for content determination of aesculetin,fraxetin,hyperoside and quercetin in Ledum palustre leaves.The reversed-phase high performance liquid chromatography(RP-HPLC) system consisted of an Inertsil ODS-SP column(5μm,4.6mm×250mm,series number: 9KI92609).Volume ratio of methanol to 0.2% acetic acid(43∶57) was used as mobile phase.Flow rate was 1.0mL·min-1,and the detector wavelength was set at 350nm.The average recoveries of aesculetin,fraxetin,hyperoside and quercetin were 99.71%,98.62%,100.13%,and 98.33% with relative standard deviations of 1.45%,1.53%,1.04% and 1.69%,respectively.The RP-HPLC method is simple,accurate and repeatable,and it could be applied to the content determination of four components in L.palustre leaves.
