新城疫病毒河北分离株F基因的克隆与序列分析

摘    要:通过RT-PCR技术扩增和克隆2003—2007年由河北省分离到的、具有一定代表性的鸡新城疫病毒分离株F基因,与GeneBank中已发表的NDV不同基因型代表株F基因相应核苷酸片段及其氨基酸序列进行比较,构建系统发育进化树并确定分离株的毒力强弱及所属基因型。结果显示,分离株在该区段没有碱基缺失和插入,但有多处点突变分散存在。分离株F基因核苷酸序列与国内外已发表毒株相应序列核苷酸同源性为80.5%~98.5%,推导的氨基酸序列同源性为77.5%~97.7%。表明目前河北新城疫的流行以基因Ⅶ型为主,同时兼有传统的基因Ⅵ型和基因Ⅱ型。

Abstract:The F gene of Newcastle disease virus isolated from Hebei between 2003-2007 were amplified by RT-PCR. Gene F and their corresponding amino acid sequence of the NDV strains were compared and analyzed. The phylogenetic tree system was constructed,then the virulence and genotype of strains were confirmed. The result showed that none of the F genes of the isolates suffered any base deletion and insertion,but there were mutations in many points scattering about. The homologous rate of F gene nucleotides sequences was 80.5%~98.5% with that of related sequences reported home and abroad. The amino acid residues predicted shared an identity between77.5%~97.7% with that of other strains. The results revealed that the NDV epidemic belonged to 3 genotypes (II,VI and VII) and the genotype VII NDV was the major pathogeny causing the epizootic of ND in Hebei.
