‘Siberia’百合再生体系的建立

摘    要:以东方百合‘Siberia’离体鳞片为外植体,研究不同激素质量浓度对芽的诱导、增殖、小鳞茎形成、叶片和叶柄再生及生根的影响。结果显示:最适不定芽诱导培养基为MS+6-BA 2.0mg/L+NAA 0.5mg/L,分化率85%;最适增殖培养基为MS+6-BA 0.7mg/L+NAA 0.2mg/L,芽增值系数为3.83;不定芽形成鳞茎的适宜培养基为:MS+6-BA 0.2mg/L+NAA 0.5mg/L+7%蔗糖,增值系数2.47。鳞片叶片的最适再生培养基为MS+6-BA 0.5mg/L+2,4-D 1.0mg/L,分化率为92.5%;叶柄的最佳再生培养基为MS+6-BA 1.0mg/L+NAA 0.5mg/L,分化率为83.3%;暗培养对鳞片叶片及叶柄的再生无显著性的差异,分化率均在80%以上;最适生根培养基为1/2MS+NAA 0.2mg/L,生根率100%。

Abstract:The bulb scales of the oriental lily 'Siberia' were treated as initial explants to test the inducement of bud,the bulb formation,the regeneration of leaf and leafstalk and rooting under different hormone concertrations.The result indicated that the optimal medium for adventitious bud came to be MS+6-BA 2.0 mg/L+NAA 0.5 mg/L with the differentiation rate of 85%.The most appropriate culture medium for multiplication was MS+6-BA 0.7 mg/L+NAA 0.2 mg/L,and the multiplication coefficient of bud was 3.83.The best transformation medium from adventitious bud to bulb was MS+6-BA 0.2 mg/L+NAA 0.5 mg/L+7% sugar,and the multiplication coefficient was 2.47.The most appropriate regeneration medium of scale,leaf was MS+6-BA 0.5 mg/L+2.4-D 1.0 mg/L,with the differentiation rate of 92.5%.And the culture medium MS+6-BA 1.0 mg/L+NAA 0.5 mg/L was the regeneration of leafstalk the most proper with the differentiation rate of 83.3%.Dark training showed no obvious difference on the regeneration of bulb scale,leaf and leafstalk,for the differentiation rates all came to be over 80%.And finally,the most appropriate culture medium for rooting was 1/2MS+NAA 0.2 mg/L,all of them were rooted under the treatment.
