部分枣属植物硅胶干燥叶片DNA提取方法的比较

摘    要:以硅胶干燥15 D和半年的3种野生枣属植物叶片为试材,分别采用简易CTAB法、高盐沉淀法和高盐低PH法3种方法提取基因组总DNA,通过A260/A280、琼脂糖凝胶电泳、PCR扩增分析对提取的基因组DNA进行了鉴定。半年内不同保存时期的材料对于DNA提取没有明显差异;高盐沉淀法所得DNA纯度最高。

Abstract:Improved CTAB,high salt precipitation,and high salt and low pH method were employed to extract genomic DNA from the leaves of 3 species of Ziziphus Mill.saved in silica for 15 days and for half a year.The extracted DNA was tested using the value of A_(260)/A_(280),agarose gel electrophoresis and PCR amplification.The DNA from the leaves saved in silica for half a year is good for PCR amplification and the method of high salt precipitation is best.
