| 摘 要： | 研究了不同玻璃化液、冷冻基础液以及卵母细胞的成熟阶段等因素对山羊卵母细胞OPS法玻璃化冷冻后发育效果的影响。结果表明,玻璃化液20%EG+20%DMSO和EDFS40对卵母细胞的冷冻保护效果间差异不显著。与成熟卵母细胞相比,未成熟卵母细胞对低温更敏感,使用现有的冷冻方法和冷冻保护液冷冻保存未成熟的山羊卵母细胞尚无法得到满意的结果。DPBS和M199均可作为配制冷冻保护液的基础液。

| Abstract: | The factors of affecting the developmental capacity of goat oocytes after open pulled straw vitrification,including different vitrification solutions,media and in vitro matured stage are all studied.The results indicate that the cryoprotection of oocytes using 20% ethylene glycol and 20% dimethylsulphoxide and EDFS40 is the same.Cryopreserving goat immature oocytes with the freezing methods and cryoprotectants now can not get satisfactory results,because the immature oocytes are more sensitive than the mature oocytes.Both Dulbecco's phosphate-buffered saline and medium 199 can be used for confecting cryoprotectants as media. |
