玉米大斑病菌犬尿氨酸途径关键酶基因的鉴定

摘    要:犬尿氨酸途径(Kynurenine pathway,KP)是色氨酸分解代谢的主要途径,其途径关键酶基因参与多种重要的生物过程。为明确玉米大斑病菌犬尿氨酸途径及其途径关键酶基因在病菌生长发育和致病过程中的功能,本研究利用生物信息学方法,对玉米大斑病菌犬尿氨酸途径中的关键酶基因进行鉴定;利用玉米大斑病菌的表达谱数据,对犬尿氨酸途径关键酶基因在病菌生长发育和侵染过程中的表达水平进行分析。结果发现,玉米大斑病菌中含有犬尿氨酸途径关键酶犬尿氨酸单加氧酶(KMO)、犬尿氨酸酶(KYN)、犬尿氨酸氨基转移酶(KAT)和吲哚-2, 3-双加氧酶(IDO)的编码基因,犬尿氨酸途径关键酶基因在病菌生长发育和侵染的不同时期呈现高表达水平,表明玉米大斑病菌中存在犬尿氨酸途径,犬尿氨酸途径关键酶基因可能在病菌生长发育和侵染过程中具有重要功能。研究结果为阐明玉米大斑病菌犬尿氨酸途径在生长发育和致病过程中的功能奠定了基础。

Abstract:The kynurenine pathway is a major pathway for tryptophan catabolism, and the key enzymes are involved in many important biological processes. In order to clarify the functions of the kynurenine pathway and its key enzymes in the growth, development and pathogenicity of Setosphaeria turcica, bioinformatics methods were used to identify the key enzymes in the kynurenine pathway of Setosphaeria turcica. The expression levels of the key enzymes of the kynurenine pathway in the growth, development and infection of Setosphaeria turcica were analyzed using the expression profiles of Setosphaeria turcica. The results showed that the genes encoding kynurenine monooxygenase(KMO), kynurenine monooxygenase(KYN), kynurenine aminotransferase(KAT) and indole-2,3-dioxygenase(IDO), which are key enzymes of the kynurenine pathway, were expressed at high levels during the growth, development and infection of Setosphaeria turcica. These results suggested the existence of the kynurenine pathway in Setosphaeria turcica and key enzymes of kynurenine pathway may play important function in the growth,development and pathogenicity of Setosphaeria turcica. These results lay a foundation for elucidating the functions of the kynurenine pathway in the growth, development and pathogenicity of Setosphaeria turcica.
