小麦醇溶蛋白A-PAGE电泳鉴定技术的分析

摘    要:为了进一步对小麦醇溶蛋白A-PAGE电泳的鉴定方法进行标准化,比较了凝胶浓度、缓冲体系及其pH值对电泳分辨率的影响,并初步筛选出了适合作为醇溶蛋白电泳标准样品的国内小麦品种。结果表明,丙烯酰胺和N,N-甲叉双丙烯酰胺2个单体的交联百分浓度(C)为4.0%,总百分浓度(T)为6%,缓冲体系为甲酸-甘氨酸(pH3.0)混合体系的A-PAGE电泳效果较好;通过国内40个小麦品种与加拿大硬红春麦Marquis的R24、R50、R79标准条带相比较,确定荔丰3号可以作为国内小麦醇溶蛋白电泳的标准样品。

Abstract:To standardize the method of wheat gliadin A-PAGE and find the native wheat varieties(lines) which can be used as the standard samples of electropherogram.The influences of the gel concentration,buffer system and its pH on the electrophesis' isolation capacity were analyzed.The results showed that the covalent percentage concentration(C) was 4.0%,the total percentage concentration of arc to bis(T) was 6%,and the formate-glycine mix buffer(pH 3.0) had the sound effects.Compared with the Canadian spring wheat Marquis' R24,R50 and R79 bands,the"Lifeng NO.3"cultivar could be chosen from 40 native wheat varieties as the wheat gliadin standard sample.
