羊绒生长期、休止期陕北白绒山羊皮肤组织中KAP9.2和Hoxc13基因的表达

摘    要:【目的】研究角蛋白相关蛋白9.2基因(KAP9.2)和Hoxc13基因在羊绒生长不同阶段皮肤组织中的表达量对产绒量的影响,为羊绒生长的调控机理研究奠定基础。【方法】以陕北白绒山羊为研究对象,以奶山羊作为对照,利用qRT-PCR方法,检测羊绒生长期和休止期高产、低产绒量陕北白绒山羊皮肤组织中KAP9.2和Hoxc13基因的表达量。【结果】 对于KAP9.2基因,在羊绒生长期高产绒量陕北白绒山羊的相对表达量极显著低于低产绒量陕北白绒山羊(P<0.01),而在羊绒休止期高产绒量陕北白绒山羊的相对表达量却显著高于低产绒量陕北白绒山羊和奶山羊(P<0.05);对于Hoxc13基因,在羊绒休止期高产和低产绒量陕北白绒山羊的相对表达量极显著高于奶山羊(P<0.01)。高产和低产绒量陕北白绒山羊皮肤组织中KAP9.2和Hoxc13基因在生长期、休止期平均相对表达量的变化趋势相似,均表现为生长期极显著低于休止期(P<0.01)。【结论】KAP9.2对绒毛生长有抑制作用;Hoxc13对KAP9.2基因的表达可能具有调控作用。

Abstract:【Objective】 The objective of the present study was to detect the relative expression of KAP9.2 and Hoxc13 gene in anagen and telogen of Shaanbei white cashmere goat,and to investigate their influence on cashmere yield.【Method】 qRT-PCR was used to detect the expression levels of KAP9.2 and Hoxc13 gene with high cashmere yield and low cashmere yield in anagen and telogen of Shaanbei white cashmere goat,with dairy goat as control.【Result】 The relative expression of KAP9.2 with high cashmere yield was extremely significantly lower than that of low cashmere yield goats in anagen(P<0.01),while in telogen the relative expression of high cashmere yield individuals was significantly higher than that of low cashmere yield and dairy goats(P<0.05);As to Hoxc13 gene,the relative expression with high and low cashmere yield goats was also extremely significantly higher than that of dairy goats in telogen(P<0.01).The result showed that the average relative expression of KAP9.2 and Hoxc13 gene with high cashmere yield and low cashmere yield had similar varying tendency in anagen and telogen,the relative expression in anagen was extremely significantly lower than that of telogen(P<0.01).【Conclusion】 The results indicated that KAP9.2 gene inhibits the growth of cashmere,and Hoxc13 gene might have important role in regulating the expression of KAP9.2 gene.
