摘    要:利用已经连续培养 35~ 38代、其腋芽增殖生长能力已衰退的香椿试管苗为材料 ,研究了Thidiazuron(TDZ)对叶、叶柄、茎段外植体形成愈伤组织的反应以及对腋芽增殖生长、生根的影响 ,结果显示 ,TDZ+0 .2 m g/ L NAA能使不同外植体形成愈伤组织的能力显著提高 ,其中茎段形成愈伤组织的能力较强。 TDZ小于0 .0 2 mg/ L时对芽的增殖生长起促进作用 ,而且可恢复腋芽旺盛的分枝增殖能力 ;TDZ高于 0 .0 2 mg/ L 则全面抑制生长 ,并随质量浓度的升高变态苗增多 ;经 TDZ培养过的试管苗其生根率并未降低。

Abstract:The experiment was carried out upon the shoots of Toona sinensis which had degenerated on qualities of growth and proliferation after being subcultured 35-38 generations.The effects of Thidazuron (TDZ) on callus inducement,proliferation and rooting have been studied.The results show that the quality of callus inducement would be increased obviously when supplemented with TDZ and 0.2 mg/L NAA in the MS medium,and stem explants would induce callus more easily than leaves and leaf stalks.The results revealed that TDZ improved proliferation and growth of shoots when its concentration was lower than 0.02 mg/L,proliferation and growth of the shoots have been recovered from the degeneration.But when its concentration was above 0.02 mg/L,they were restrained completely,at same time,there were more abnormal shoots with increasing concentration in medium.The rooting rate of plantlets has not been decreased by subcultured in the medium which supplemented with TDZ.
