奶牛胚胎直接投入液氮冷冻保存试验

将奶牛7日龄胚胎,常温下在含128.9g/L甘油PBSS肉孵育20min后移入193.2g/L甘油+85.6g/L蔗糖PBSS内孵育7min,装管后即投入液氮内冷冻保存。182d后解冻8枚,用含342.3g/L蔗糖PBSS脱去甘油,7枚可供移植用,胚胎可用率为87.5％(7/8),移植给7头受体牛,3头妊娠,妊娠率为42.9％。

A Preliminary Study on the Cryopreservation of Bovine Embryos by Direct Plunge into Liquid Nitrogen

Some bovine embryos at the age of 7 days were incubated for 20 minutes in PBSS solution with 128. 9 g/L glycerol at room temperature, then placed in PBSS containing 193. 2 g/L glycerol and 85. 6 g/L sucrose for 7 minutes, and finally transferred into straw. The straws with embryos were directly plunged into liquid nitrogen for cry-opreservation. After 182 days of preservation, 8 embryos were unfrozen and washed with PBSS solution containing 342. 3g sucrose to remove the glycerol from them. Seven embryos were used for transferring, and the embryo recovery rate was 87. 5%. The embryos were then transferred into 7 cow recipients, 3 of which became pregnant, and the pregnacy rate was 42. 9%.
