葡萄无核基因RAPD标记的序列分析

摘    要:报道了用QiagenKit纯化RAPD遗传标记,用细菌质粒M13克隆RAPD标记的DNA片段,采用自动荧光DNA测序仪(型号373A)测定了该DNA片段的核苷酸组成及其排列顺序。无核白RAPD标记由519对核苷酸及其特定序列组成。因为无核白是无核基因的原始供体,作者认为所获无核白RAPD标记的DNA序列,可以作为合成探针的基础,用于检测无核葡萄育种和无核品种的无核性。

Abstract:This paper presents the sequence of RAPD marker linked to the seedless genes in grapes. The isolation DNA was purified by Qiagen kit and inserted into plasmid PCR TM II(Invitrogene). The sequencing was performed based upon automated flourescent DNA sequencer systems (Model 373A). The marker from the seedless originator Thompson Seedless consists of 519 bp with specific nucleotide sequence. Because of the initial donors of seedless genes from Thompson Seedless, it is suggested that the sequence of Thompson Seedless might be used as basis for synthesizing the probe for detecting seedlessness both in seedless breeding and identification of seedless varieties from it.
