甘农2号杂花苜蓿愈伤组织诱导及体细胞胚和芽分化的研究

摘    要:以甘农2号杂花苜蓿下胚轴为材料,MS为基本培养基,研究了不同浓度的2,4-D、NAA、KT、BA、蔗糖及2,4-D作用时间对甘农2号杂花苜蓿愈伤组织的诱导、体细胞胚和芽分化的影响.结果表明,3 mg/L 2,4-D+1.5 mg/L NAA为愈伤组织诱导阶段最佳激素组合;2,4-D作用时间以15 d为宜;最佳的芽分化培养基为MS+0.5 mg/L KT+0.1 mg/L NAA+7 g/L琼脂+20 g/L蔗糖.

Abstract:Taking hypocotyls of Medicago variaGannong No.2 as the explants and MS as basic medium,the effects of different addition concentration of 2,4-D,NAA,KT,BA,sucrose and the inducing time of callus on the callus induction,somatic embryogenesis and shoot differentiation were studied.The result showed that 3 mg/L 2,4-D 1.5 mg/L NAA was the optimum hormone combination,the proper action time of 2,4-D was 15 days,and the optimum culture medium for shoot differentiation was MS 0.5 mg/L KT 0.1 mg/L NAA supplemented with 7 g/L agar and 2 g/L sucrose.
