嗜热脂肪芽孢杆菌中普鲁兰酶的分离纯化

摘    要:嗜热脂肪芽孢杆菌经细胞破碎机破壁后,采用硫酸铵盐析法除杂蛋白,DEAE-32阴离子交换纤维素纯化胞内普鲁兰酶,并通过L9(34)正交试验优化盐析除杂蛋白工艺条件.结果表明:菌体与缓冲液比1:3、硫酸铵饱和度40%、pH值7.0分离效果最佳,其中,pH值对分离效果的影响差异性显著(F>F0.05);纯化后的普鲁兰酶比活力...

Abstract:Bacillus stearothermophilus was broken using cell crushing machine,the impurity protein was taken out using ammonium sulfate salting out method.Cell pullulanase was purified using DEAE-32 cellulose anion-exchange.The salting out conditions were optimized through orthogonal test.The results showed that the separate condition was the best when the buffer was three times of volume than bacteria,ammonium sulfate saturation was 40%,pH value was 7.0.pH value had an great effect on the separation,differences were ...
