培养基和培养条件对葡萄试管苗生根的影响

摘    要:以矢芙罗莎等4种葡萄试管苗为材料,研究了蔗糖浓度、琼脂浓度、pH、光强及温度对葡萄试管苗生根的影响.结果表明:蔗糖浓度为15~20 g/L对葡萄试管苗的生根最有利;pH值对试管苗平均根系数量、根粗、根长和根冠比影响显著,生根数与平均根长随pH值升高而减小,平均根粗与根冠比随pH值的增大而增大;琼脂浓度显著影响葡萄试管苗生根,随着琼脂浓度的增加,根源基产生时间延迟,平均根长和根冠比降低,而平均根粗和生根数增加;生根数在3种温度条件下无显著差异,平均根粗随温度的升高略有下降,平均根长和根冠比随培养温度的升高而增加,在35℃时降低;平均生根数、根长、根冠比和根粗均随光照强度的增强而增大.

Abstract:Effects of different culture conditions including different concentration of sucrose and agar added in media,different pH values, culture temperature and light intension on rooting in vitro of test-tube plantlets of Vitis vinifera L.were carried out.The results showed that the optimum concentration of sucrose for rooting was 15~20 g/L.pH value had significant effects on root number,root diameter,root length and root-shoot ratio of test-tube plantlets cultured with different media,pH value increased,root number and average root length decreased,root diameter and root-shoot ratio increased.Concentration of agar significantly affected the rooting of test-tube plantlets,the high agar concentration was the later root primordium produced,meanwhile,as concentration of agar increased,average root length and root-shoot ratio decreased,average root diameter and root number increased.Root number showed no significant differences among three temperature treatments,while the temperature increased,average root diameter decreased a little,average root length and root-shoot ratio increased first,but when temperature reached to 35 ℃,they decreased.With the increasing of light intension,number,length and diameter of root and root-shoot ratio were all increased.
