摘    要:构建了单子叶植物双价表达载体pBI121-Ubi-Bar/Chi,其中含有木霉几丁质酶基因Chi和抗除草剂Bar基因,均由玉米Ubi启动子驱动.以烟草无菌苗叶片为受体,用农杆菌介导法将目的基因转入烟草中;以Bar基因作为选择标记,PPT为选择剂进行筛选,获得了一定数量的转基因植株.结果表明:经PCR分析,初步确定目的基因已经整合到烟草基因组中.

Abstract:Monocotyledon plant bivalent expression vector was constructed as pBI121-Ubi-Bar/Chi which included Trichoderma chitinase gene Chi and resistant herbicide Bar gene that were started by Ubi promoter of maize.The target genes were transferred directly to tobacco by Agrobacterium mediated method,and the receptor was leaf of tobaco aseptic seedling.Some trans-genetic plants were obtained by selecting with the PPT as selector, and Bar gene was selecting marker. The target genes had been integrated to tobacco genome preliminarily through PCR analysis.
