川芎多糖的分离纯化及其单糖组成测定

摘    要:为了解川芎多糖的分子量和单糖组成,为药量学研究和中成药加工奠定基础,采用水提醇沉法提取川芎多糖,DEAE-纤维素柱层析法纯化,高效凝胶渗透色谱法测定分子量。然后用硫酸水解,1-苯基-3-甲基-5-吡唑啉酮衍生水解后的单糖,衍生物采用高效液相色谱法测定多糖的单糖组成。结果表明,DEAE-纤维素柱层析获得3个川芎多糖组分LCXP-1、LCXP-2和LCXP-3,分子量分别为11.916、20.793和701.875kDa。LCXP-1和LCXP-2均由甘露糖、葡萄糖、半乳糖、阿拉伯糖组成,摩尔比分别为1:495:3.7:1.2和1:632:4.3:1.2;LCXP-3由甘露糖、鼠李糖、半乳糖醛酸、葡萄糖、半乳糖、阿拉伯糖组成,摩尔比为1:6.5:1.5:248:50:14。该方法准确、灵敏度高,适用于川芎多糖中单糖组成的测定。

Abstract:To study the monosaccharide compositions and molecular weights of Ligusticum chuanxiong polysaccharide,and lay the foundation for pharmacometry and processing of proprietary Chinese medicine,the polysaccharide was extracted by hot water and precipitated by alcohol.The product was purified by DEAE-Cellulose,molecular weights were determined by high performance gel permeation chromatography(HPGPC) and the monosaccharide compositions were determined by HPLC using precolumn derivatization with 1-phenyl-3-methyl-5-pyrazolone(PMP).The results showed that three polysaccharides(LCXP-1,LCXP-2 and LCXP-3) were obtained,molecular weights were 11.916,20.793 and 701.875 kDa,respectively.Both LCXP-1 and LCXP-2 were composed of mannose,glucose,galactose and arabinose in molar ratio of 1∶495∶3.7∶1.2 and 1∶632∶4.3∶1.2;LCXP-3 was composed of mannose,rhamnose,galacturonic acid,glucose,galactse and arabinose in molar ratio of 1∶6.5∶1.5∶248∶50∶14.The method is accurate and sensitive,thus suitable for the analysis of monosaccharide composition of Ligusticum chuanxiong polysaccharide.
