摘    要:以草地早熟禾(Poa pratensis L.)品种"午夜"的成熟种子及幼叶为外植体,MS为基本培养基,研究不同外源植物激素对其愈伤组织形成的影响。结果表明,成熟种子为最佳外植体,由成熟种子诱导出的愈伤组织的出愈率与质量均高于叶片;MS+2,4-D 5.0 mg/L培养基为成熟种子较为适宜的诱导培养基,诱导率达50%。在MS+2,4-D5.0 mg/L继代培养基中,有黄绿色、表面干燥的颗粒状愈伤组织长出。MS+2,4-D 7.0 mg/L是叶片较为适宜的诱导培养基,诱导率为34%,但长出的愈伤组织表面多水分。

Abstract:In the current study,a kind of mature seeds of kentucky bluegrass(Poa pratensis L.) named 'Midnight' was used as the explants to compare different concentrations on callus induction of bluegrass,where the MS as the basic culture medium.The data suggest that mature seeds is the best explants due to their higher rate and quality of the callus compare with those from the leaves;MS+2,4-D(2,4-dichlorophenoxyacetic acid),5.0 mg/L seems to be the best suitable culture medium for the mature seeds as the callus induction rate was 50%,and dry-faced granulated callus colored yellow green in the following generation of culture medium.Additionally,MS+2,4-D(2,4-dichlorophenoxyacetic acid),5.0 mg/L was suggested to be most suitable for leaves with a callus induction rate at 34%,although there were some washy callus was observed in herein study.
