摘    要:以不同甘蔗品种幼苗期幼嫩叶片为材料,以改良SDS法和DNA快速提取法对甘蔗基因组DNA进行提取,并以SSR-PCR检测比较两者的扩增效果。结果表明,两种方法提取获得的基因组DNA条带基本一致,条带较为清晰,重复性好且稳定,均可满足SSR-PCR扩增的需要。但DNA快速提取法简化了SDS改良法,具有高通量、操作方便、简单、快速、成本低、DNA存放时间较长等优点,能在较短时间内完成大量样品的DNA提取。

Abstract:The young leaves of different sugarcane varieties were sampled at settling stage to extract genomic DNA,and a rapid DNA extraction method was developed by modifications in SDS method.The extracted DNA was amplified in PCR using SSR primers.The results showed no differences in the amplified bands of the DNA isolated using the two different methods.The DNA isolated from rapid method showed clear and reproducible bands,and was found suitable for the application in marker related studies.Further,the DNA isolate...
