不同激素组合对番茄温敏核不育系及其杂交组合花药愈伤组织诱导效应

摘    要:【目的】研究不同激素组合与不同材料基因型互作对番茄温敏核不育系及其杂交组合花药愈伤组织诱导的效应。【方法】以番茄温敏核不育系大107、地D4及其杂交组合大107x牛番茄、地D4x红昆仑为材料,进行不同激素组合(CPPU、TDZ、KT与2,4-D+6-BA)诱导花药愈伤组织诱导试验。【结果】以MS+1.0mg/L2,4-D+0.5mg/L6-BA为基本诱导培养基,分别与不同浓度CPPU、TDZ或KT组合,对不同基因型番茄花药愈伤组织诱导具有明显促进效果,CPPU、TDZ和KT的适宜浓度分别为0.4—0.6、0.03和1.5mg/L。在相同激素条件下,番茄温敏核不育系的花药愈伤组织诱导效果明显优于其杂交组合,平均出愈率和平均愈伤组织直径大小均为:地D4〉大107〉地D4x红昆仑〉大107x牛番茄。【结论】不同基因型番茄花药对激素的敏感性不同;不同激素组合对番茄花药愈伤组织诱导的效应差异显著,以KT与2,4-D、6-BA配组对番茄温敏核不育系及其杂交组合花药愈伤组织的诱导具有显著调控作用。

Abstract:Objective]The present experiment was aimed to study the effects of different hormone combinations on callus induction in anthers taken from tomato thermo-sensitive genic male sterile lines (TSGMSLs) and their hybrid combinations. Method ]Tomato TSGMSLs, viz., Da 107 and Di D4, and their hybrid combinations, viz., Da 107xNiufanqie and Di D4xHongkunlun were used in the studies. Combinations of different hormones (CPPU, TDZ, KT) in basic MS medium (containing 1.0 mg/L 2,4-D+ 1.0 mg/L 6-BA) were used for anther callus induction. Result ] Adding different CPPU, TDZ or KT concentrations in basic MS medium significantly improved the induction of anther callus, and the suitable con- centration of CPPU, TDZ and KT was 0.4-0.6, 0.03 and 1.5 mg/L, respectively. Under the same hormone combination conditions, the anthers taken from tomato TSGMSLs showed better callus induction compared to those taken from hybrid combinations, the order of the average rate of callus induction and average diameter of callus was: Di D4〉Da 107〉Di D4x Hongkunlun〉Da 107xNiufanqie. Conclusion]Anthers of different tomato genotypes differed in their sensitivities to different hormones. Furthermore, different combinations of hormones also differed in affecting induction of tomato anther callus, out of them, the combinations of KT and 2,4-D+6-BA showed best results.
