摘    要:【目的】探讨铁皮石斛种子萌发、芽增殖和生根的适宜培养基,为铁皮石斛组织快繁技术提供依据。【方法】以铁皮石斛种子为外植体进行培养,研究6-BA(0-3.0mg/L)、NAA(0.1-0.3mg/L)、香蕉泥(15%~25%)和马铃薯泥(15%~25%)对铁皮石斛种子萌发、芽增殖和生根壮苗的影响。【结果】适宜种子萌发的较好培养基为MS+20%马铃薯泥,芽增殖最适培养基为MS+2.0mg/L6-BA,增殖率达282.7%;最适生根培养基为MS+15%香蕉泥,生根率达100%。【结论】经马铃薯泥、香蕉泥培养的铁皮石斛组培苗,生长健壮,能为生产提供大量的种苗,因而此类培养基可用于大规模生产铁皮石斛组培苗。

Abstract:Objective]The experiments were conducted to find out the suitable culture medium for the seeds germination,bud multiplication,roofing and growth of plantlet in Dendrobium candidum Wall.ex Lindl,and to develop a technique for its rapid propagation. Method]Seeds of Dendrobium candidum were used as explants in tissue culture,and the effects of different media with different concentrations of 6-BA (0-3.0 mg/L) and NAA (0.1-0.3 mg/L),supplemented with mashed banana (15-25%) or mashed potato( 15-25% ),on the seed germination,bud multiplication,rooting and growth of plantlets were investigated. Result]The most suitable culture medium for seed germination and bud multiplication was MS+20% mashed potato and MS+6-BA (2.0 mg/L),respectively. However,the best suitable medium for rooting was found MS+15% mashed banana which gave 100% rooting rate. Conclusion]The strong rooting plantlet can be obtained in medium supplemented with mashed potato and mashed banana. This medium may be recommended for the large scale production of Dendrobium candidum plantlets.
