摘    要:【目的】研究不同基因型苦瓜核糖体失活蛋白(RIPs)在种子发育过程中的累积规律,为苦瓜RIPs的进一步利用提供理论依据。【方法】选取3个不同组群苦瓜品种为材料,对不同发育时期种子的盐溶性蛋白提取物进行SDS-PAGE分析。【结果】3个苦瓜品种种子的盐溶性蛋白组分存在明显差异,其中以分子量为20.0~29.0kDa的蛋白质差异最为明显;29.0kDa分子量的蛋白质在长大果型材料MC1及密瘤小果型材料MC11中的表达量比野生型材料MC18略高。3个苦瓜材料的29.0kDa蛋白质在种子不同发育时期的表达量无明显差异,然而,在不同栽培季节其表达量差异显著。【结论】不同类型苦瓜材料种子中的RIPs含量变化在种子发育不同时期没有明显差异,而成熟种子具有产量高、易采收和贮藏等优点,是作为RIPs生产原材料采收的适宜时期;由于不同类型苦瓜的种子产量潜力有较大差异,因此,选择RIPs含量高且种子产量高的材料作为RIPs生产的品种较为适宜。

Abstract:【Objective】In order to provide theoretical information for the utilization of ribosome-inactive proteins (RIPs) in M. charantia (bitter gourd), the trends of accumulation of ribosome-inactive proteins (RIPs) during seed development stage of different M. charantia genotype were studied. 【Method】The salt-soluble proteins of three genotypes of M. charantia seeds obtained from different seed development stages were subjected to SDS-PAGE analysis.【Result】The results showed remarkable differences in proteins of three genotypes. The differences were more pronounced in proteins with molecular weight ranged from 20.0 to 29.0 kDa. The 29.0 kDa proteins were higher in MC1 (the large fruit type genotype) and MC11 (small-warty fruit type genotype) compared to MC18 (wild type). No significant differences were observed in the accumulation of 29.0 kDa proteins at different seed development stages in three genotypes, however, these proteins showed remarkable differences when the plants were grown in different seasons. 【Conclusion】The RIPs from seeds of three M. charantia genotypes did not showed significant differences at different development stages of seed. The maturing seed was characterized by high yield and was easy to harvest and store, and can be used to study the alterations in RIPs. As a source material for RIPs extraction, M. charantia genotype with high RIPs content and seed yield would be recommended.
