| 摘 要： | 根据GenBank上牛分枝杆菌（M.bovis）CFP-10、ESAT-6、MPB70的基因序列，设计并合成了3对扩增CFP-10、ESAT-6、MPB70基因的特异性引物，扩增片段大小分别是321、306、582bp。对牛分枝杆菌广西分离株MBT359进行以上3个基因的克隆、序列测定及分析，结果显示，可以从广西分离株MBT359扩增出大小与预期相符的基因片段，经测序证实，扩增出的片段即为3个目的基因片段。序列分析表明，广西分离株MBT359的CFP-10、ESAT-6、MPB70基因片段与国际标准强毒株AF2122/97相应基因核苷酸同源性为100％。该研究结果为广西牛分枝杆菌流行株主要基因测序、建立广西牛结核病流行株遗传多态性数据库奠定了基础。

| Abstract: | In the present study, three pairs of specific primers were designed and synthesized from the sequence of CFP-10, ESAT--6, MPB70 genes of M. boris obtained from GenBank. The CFP-10, ESAT-6, MPB70 genes of M. boris strain MBT359 were amplified by using the primers, cloned and sequenced. The amplified fragments of CFP-10, ESAT- 6, MPB70 genes were 321, 306 and 582bp, respectively. The sequences of these genes were compared with the M. boris strain AF2122/97 which resulted in 100% homology with the sequences of AF2122/97 strain. These results might be helpful in characterization of Guangxi M. boris strains and construction of genetic polymorphism data. |
