迟熟蕉柑茎尖嫁接砧木对比试验及病毒类病害鉴定

摘    要:分别以枳和阿尔及利亚夏橙作为砧木,以迟熟蕉柑芽作为接穗进行茎尖嫁接,结果枳砧嫁接成活率为9.1%,阿尔及利亚夏橙砧嫁接成活率为54.5%,将两种砧木嫁接苗再嫁接在枳砧上,其3年生幼树均生长正常,表明阿尔及利亚夏橙是茎尖嫁接迟熟蕉柑的优良砧木。应用指示植物鉴定方法和PER检测技术鉴定7株茎尖嫁接苗的柑橘碎叶病、衰退病、裂皮病和黄龙病,结果1、3.4、5、6号迟熟蕉柑单株不带碎叶病、衰退病、裂皮病和黄龙病,可作为繁殖无病苗木的备选材料;2、7号迟熟蕉柑单株不带碎叶病、裂皮病和黄龙病,但带有衰退病,推测衰退病毒源可能来自取芽的田间植株。

Abstract:In the present study, trifoliate orange and Algers Valencia orange were used as stocks and the buds of late-ripening Jiaogan mandarins were used as scion for shoot-tip grafting. The results showed that the survival rates of grafted seedlings using trifoliate orange rootstocks and Algers Valencia orange as stocks was 9.1% and 54.5%, respectively. The grafted seedlings from these two stocks were grafted on stock of trifoliate orange again and the 3-year-old trees grew normally, which revealed that Algers Valencia orange was the better stock for shoot-tip grafting of late-ripening Jiaogan mandarins. Moreover, citrus tatter leaf virus(CTLV), citrus tristeza virus(CTV), citrus exocortis viroid(CEV) and Huanglongbing (HLB) in seven shoot-tip grafted seedlings were identified by indicator plants and PCR technique. The results showed that the seedlings from 1^st, 3^rd, 4^th, 5^th and 6^th late-ripening Jiaogan mandarins did not have pathogens of CTLV, CTV, CEV and HLB, which could be used to develop virus-free seedlings, while the seedlings from 2^nd and 7^6h late- ripening Jiaogan mandarins had no CTLV, CEV and HLB, but had the pathogen of CTV that might have been come from the buds of late-ripening Jiaogan mandarins in field.
