摘    要:对85株副溶血弧菌的Ⅵ型分泌系统(typeⅥsecretion system, T6SS)相关基因的携带情况进行分析,并选取部分副溶血弧菌菌株进行混合培养,探究其种内竞争情况。利用qPCR技术,分析混合培养后T6SS相关基因的表达情况。结果表明:85株副溶血弧菌均携带T6SS2,62株副溶血弧菌携带T6SS1,其中临床分离株41株,占全部临床分离株的93.2%;环境分离株21株,占全部临床分离株的51.2%,并且T6SS1基因簇在不同来源的副溶血弧菌中存在一定的差异性。在混合培养条件下,部分T6SS1~+的副溶血弧菌存在明显的种内竞争优势。基因表达分析结果显示,较单一培养的菌株,混合培养中副溶血弧菌T6SS1和T6SS2相关基因的表达量均有明显的上调。导致这一现象的原因可能是在混合培养的条件下,副溶血弧菌可通过增强自身T6SS1和T6SS2相关基因的表达,以提升其在种内的竞争力。初步探究了副溶血弧菌Ⅵ型分泌系统对其种内竞争的影响,为副溶血弧菌T6SS功能的进一步揭示提供科学的参考。

Abstract:Vibrio parahaemolyticus type VI secretory system (T6SS) has close association with its ability to adapt to the external environment, while its influence on Vibrio parahaemolyticus intraspecific competition remains unknown. The 85 strains of Vibrio parahaemolyticus harboring the T6SS related genes was investigated, in this study. The T6SS1+ and T6SS1- strains chosen were conducted in mixed culture to explore their intraspecific competition. Meanwhile, qPCR technology was employed to analyze the expression of related genes of T6SS. The results show that although all Vibrio parahaemolyticus harbored T6SS2, T6SS1 was found in a certain number of strains. 85 strains of Vibrio parahaemolyticus all carried T6SS2, and 62 strains of Vibrio parahaemolyticus carried T6SS1, of which 41 were clinical isolates, accounting for 93.2% of all clinical isolates. There were 21 environmental isolates, accounting for 51.2% of all environmental isolates. Moreover, T6SS1 gene clusters were different in Vibrio parahaemolyticus from different sources. Most T6SS1+ strains had significant intraspecific competitive advantages, which could suppress the growth of other strain in mixed culture. Compared with mono culture strains, the expression levels of T6SS1 and T6SS2 strains were significantly up-regulated in mixed culture. The reason for this phenomenon might be that strainscould strengthen the expression of its related T6SS1 and T6SS2 genes to enhance its competitiveness in the species. This study preliminarily explored the influence of Vibrio parahaemolyticus type VI secrete system on its intraspecies competition, providing scientific reference for further revealing the Vibrio parahaemolyticus T6SS function.
