马尾藻褐藻多糖硫酸酯的分离纯化及结构研究

摘    要:为研究马尾藻Sargassum褐藻多糖硫酸酯的组成及结构,以马尾藻褐藻多糖硫酸酯粗品为原料,利用DEAE-Sepharose Fast Flow弱阴离子交换层析对粗品进行分离纯化,采用高效液相色谱法(HPLC)对粗品(F)及纯化后的两个组分(F0、F1)进行了单糖组成分析,同时利用红外光谱法、核磁共振波谱法研究了F0及F1的多糖结构。结果表明:用高效液相色谱法对单糖衍生物的分离效果良好,F、F0、F1均含有甘露糖、鼠李糖、葡萄糖醛酸、葡萄糖、半乳糖、木糖、岩藻糖;红外光谱分析显示,F和F0的硫酸基主要连接于岩藻糖的C2和C3位置。本研究结果可为马尾藻褐藻多糖硫酸酯的生物活性研究奠定基础。

Purification and structure of fucoidan from Sargasso weed Sargassum
Abstract:The fucoidan in Sargasso weed Sargassum was purified by DEAE-Sepharose Fast Flow anion-exchange chromatography and the structure characteristics of the fucoidan fractions (F0 and F1) in the crude fucoidan (F) were analyzed by infrared spectrum ( IR) , high performance liquid chromatography ( HPLC) and nuclear magnetic resonance ( NMR) spectroscopy to evaluate the monosarccharide composition and structure of fucoidan from Sargas-so weed. The HPLC revealed that monosaccharide composition of these fractions were comprised of mannose, glu-curonic acid, galactose, and fucose, with a little amount of rhamnose, glucose and xylose. The IR revealed that the sulfate site of F and F0 were located at Fuc C2 and C3 . The findings provide basic data for the biological activity studies of the fucoidan from Sargasso weed.
