摘    要:根据GeneBank上已发表的鹅细小病毒(GPV)基因序列,设计合成1对引物,以GPV阳性毒株鹅胚尿囊液及攻毒后死亡的雏鹅组织,提取DNA并进行PCR,结果产物扩增出的片段与预期片段440 bp大小相符,测定序列与GeneBank上GPV B株同源性达99%。试验结果显示用该方法可以特异性地诊断出GPV,比琼脂扩散试验(AGP)敏感性高,可检测最低浓度为102.5GELD50/0.2mL;检测攻毒后死亡的雏鹅组织,在脑、肝、肺、肾、肠等组织中均有GPV的存在,可以用于临床可疑病毒的分离鉴定。所建立的PCR方法具有高度的特异性、敏感性和可重复性。
