摘    要:以大活的嫩茎为外植体,进行了大活组织培养及再生体系建立的研究。试验结果表明:MS+6-BA 0.5 mg/L+2,4-D 2.0 mg/L是愈伤组织诱导和继代增殖的理想培养基;在光照条件下MS+ZT 0.6 mg/L+NAA 0.1 mg/L是愈伤组织分化的理想培养基;1/4MS+IAA 0.5 mg/L是不定芽生根和试管苗生根继代增殖的理想培养基。试管苗移栽成活率为92.1%,定植后成活率为96.8%。

Abstract:Taking the tender stems of Ranunculus sceleratus as explants,the tissue culture and establishment of regeneration system in vitro of Ranunculus sceleratus were studied.The optimum medium for callus inducement and subculture proliferation was MS+6-BA 0.5 mg/L+2,4-D 2.0 mg/L;under the condition of illumination,the optimum medium for callus differentiation was MS+ZT 0.6 mg/L+NAA 0.1 mg/L;the optimum medium for the rooting of adventitious buds and subculture proliferation of test-tube seedlings was 1/4 MS+ IAA 0.5 mg/L.The transplanting survival rate of the test-tube seedlings was 92.1%,and the stable planting survival rate was 96.8%.
