百合农杆菌介导的遗传转化受体系统的建立

摘    要:以东方百合品种"西伯利亚"作为研究材料建立了百合农杆菌介导的受体系统。鳞片块不定芽的分化以MS培养基中添加0.5mg/L2,4-D和0.5mg/LBA为最好;试管苗小叶块和小鳞茎块的不定芽分化均以MS培养基中添加1.0mg/LBA和0.05mg/LIAA为最好;卡那霉素的筛选浓度小叶块确定为100mg/L,而小鳞茎块确定为125mg/L;羧苄青霉素抑菌浓度确定为300~400mg/L。

Establishment of Acceptor System of Agrobacterium-mediated Transformation of Lily
Abstract:The acceptor system of the agrobacterium-mediated transformation of lily was established by using Lilium seberia as testing material. MS medium supplemented with 2,4-D 0.5 mg/L and BA 0.5 mg/L was the best for the differentiation of adventitious bud from the scale. The differentiation of adventitious bud from the leaf piece and bulbil piece of the plantlets in vitro was the best on MS medium supplemented with BA 1.0 mg/L and IAA 0.05 mg/L. The screening concentration of Kanamycin for the leaf piece was 100 mg/L and for the bulbil piece was 125mg/L. The proper concentration of Carbenicillin for restraining bacteria was 300~400mg/L.
