镉对姬松茸细胞悬浮培养的影响及其在细胞中的分布

摘    要:主要研究姬松茸细胞液体培养、亚细胞操作、镉对细胞生长的影响和镉在细胞中的分布。结果表明,适宜培养基为5%麸皮、1%蔗糖、1%葡萄糖、蛋白胨、0.2%KH2PO4、0.1%MgSO4;培养基中低浓度镉能促进细胞的生长;姬松茸细胞能大量积累镉,而且镉大部分积累在细胞壁上(占83.2%),少量进入细胞质、液泡中;最好的细胞壁分解酶为溶壁酶和纤维素酶,原生质数可达4.6^*10^6个/mL。

Abstract:The paper deals with the suspension culture of Agaricus Blazei Murrill Cells, sub-cell manipulation,the effects of cadmium on the growth of the cell and the distribution of cadmium. The experimeatal results showed that the suitable cultural medium was 5% wheat bran, 1% sugar, 1% glucose, 0.2% peptone, 0.2% KH 2PO 4?0.1%MgSO 4. Low concentration of cadmium was good to the growth of cells. The Agaricus Blazei Murrill cell could accumulate a lot of cadmium from cultural medium. Most of the cadmium distributed in cell wall, little in plasma and vacuum. The best enzymes to discompose cell wall were Lywallzyme and cellulase. The number of plasma could reach as much as 4.6 *10 6 per milliliter.
