大豆蛋白酶解物的抗癌活性研究

摘    要:以大豆分离蛋白为原料,用中性蛋白酶、木瓜蛋白酶、碱性蛋白酶和胃蛋白酶分别对其进行酶解,得到具有抗癌活性的酶解物,测定大豆分离蛋白的水解度和酶解物对胃癌细胞的生长抑制率。结果表明,木瓜蛋白酶酶解物的癌细胞生长抑制率为27.36%,显著高于其他三种酶解物的抗癌活性,其次是中性蛋白酶(21.20%)和碱性蛋白酶(18.01%),酶解物抗癌活性最低的是胃蛋白酶(11.20%)。四种蛋白酶在最适水解条件下作用于大豆分离蛋白时,水解度的排列次序为:碱性蛋白酶中性蛋白酶木瓜蛋白酶胃蛋白酶。大豆分离蛋白的水解度与酶解物的细胞生长抑制率不呈线性关系。

Abstract:Soy protein isolate was treated as raw material.The hydrolysates with anti-cancer activities were prepared by enzymolysis separately with neutral protease,papain,alkaline protease and pepsin.The hydrolysis degrees and cancer cell growth inhibitory values of hydrolysates were detected.The results showed that the anti-cancer activity of the papain hydrolysate was 27.36%,higher than those of other protease hydrolysates,remarkably.The followed ones were neutrase hydrolysate(21.20%) and Alcalase hydrolysate(18.01%).The pepsin hydrolysate had the lowest cancer cell growth inhibitory value(11.20%).On the optimum enzymolysis conditions,the hydrolysis degree order of the soybean protein was like this: alkalase>neutrase>papain>pepsin.There was no linear relationships between cancer cell growth inhibitory values and the hydrolysis degrees.
