农杆菌介导iaaM基因黄瓜遗传转化体系的建立

摘    要:以已经建立的高频黄瓜再生体系为基础,从影响黄瓜转基因体系的农杆菌侵染时间,共培养时是否加入乙酰丁香酮等因素优化了黄瓜遗传转化体系;将单性结实基因iaaM以农杆菌介导法导入到东农649品种黄瓜子叶节中,经过不定芽诱导和生根等过程获得了转基因株系,通过特异性引物的PCR鉴定,40个株系扩增出iaaM目的基因片段。PCR-Southern检测为阳性。转基因阳性植株移栽到温室中,其果实经检测80%为单性结实黄瓜。

Abstract:On the basis of established cucumber regeneration system,transformation was established by studying factors such as Agrobacterium infection time and adding acetosyringone or not etc.The iaaM gene was transferred into the cotyledons of the cucumber of "DongNong 649" by Agrobacterium tumefaciens and the transformed cucumber lines were obtained through the process of shooting and rooting.Through the PCR detection 40 lines showed the existence of the iaaM gene fragment.The PCR-Southern detection showed positive.Plants of transgenic cucumber was transplant to greenhouse and 90% fruit was agamogenetic cucumber by detection.
