虹鳟(Oncorhynchus mykiss)多倍体的倍性鉴定

采用红细胞核体积测量法检测虹鳟多倍体的倍性,并用流式细胞术对其准确性进行验证,统计二倍体、三倍体与四倍体的核质比和微核率。结果显示,红细胞核体积测量法检测的三倍体诱导组(109尾)中有81尾三倍体,四倍体诱导组(154尾)中有26尾四倍体;流式细胞术鉴定结果显示,三倍体诱导组(109尾)中有79尾三倍体,四倍体诱导组(154尾)中有25尾四倍体,并且全部是红细胞核体积测量法所确定的三倍体和四倍体,测量法的准确率分别达到97.5%和96.2%;二倍体、三倍体和四倍体的核体积分别为51.87±6.79,78.32±2.72,99.07±9.85μm3,质体积分别为614.88±50.77、1 088.33±93.97、1 415.72±507.55!m3,核质比分别为0.084±0.024、0.072±0.014、0.070±0.010,统计表明三者的核质比无显著差异(P>0.05);三个倍性红细胞均未发现微核现象;三倍体和四倍体血液中有细胞核形态异常的红细胞,其出现率分别为0.07%和0.08%,二者无显著差异(P>0.05)。

Ploidy identification of polyploid rainbow trout (Oncorhynchus mykiss)

Using the plethysmometry of erythrocyte nucleus to identify the polyploid of rainbow trout,evaluated the accuracy using flow cytometry and did statistics of nuclear-cytoplasmic ratio and micronuclear rate.The results indicated that there were 81 tails triploid rainbow trouts in triplid induced groups measured by plethysmometry of erythrocyte nucleus and the number was 26 in tetraploid induced groups.The results of flow cytometry showed that there were 79 tails triploid rainbow trout in triploid induced groups and 25 in tetraploid induced groups,and these triploid and tetraploid rainbow trouts were all confirmative by the plethysmometry of erythrocyte nucleus.The accuracy of plethysmometry of erythrocyte nucleus reached at 97.5% and 96.2% respec-tively;the nucleus volume of diploid,triploid and tetraploid raibow trout was 51.87±6.79,78.32±2.72 and 99.07±9.85 μm3 and the volume of cytoplasm was 614.88±50.77,1 088.33±93.97 and 1 415.72±507.55 μm3 respectively,the nuclear-cytoplasmic ratio was 0.084±0.024,0.072±0.014 and 0.070±0.010 for diploid,triploid and tetraploid rainbow trout respectively.Statistics showed that there were no significant differences among three different ploidy rainbow trout in their nuclear-cytoplasmic ratio(P>0.05);no micronucleus in three different ploidy rainbow trout;erythrocyte with dyskaryosis appeared in both triploid and tetraploid bloods,and the frequency arrived at 0.07% and 0.08%,there were no significant differences(P>0.05) between them.
