甜菜基因组DNA的提取及Southern杂交分析

摘    要:分别采用常规CTAB法和SDS法提取甜菜基因组DNA,并将提取的DNA应用于Southern杂交分析,发现基因组DNA的质量对杂交信号有非常大的影响。常规CTAB法提取的DNA产生的Southern杂交信号,仅出现在高分子质量位置;SDS法提取的DNA产生的Southern杂交信号,在高分子质量和低分子质量位置均出现。SDS法提取的甜菜基因组DNA适于Southern杂交分析。

Abstract:Genomic DNA was extracted from sugar beet using the conventional CTAB method and SDS method. The quality of DNA had a great effect on the hybridization signals. The signals that were produced by genomic DNA isolated through conventional CTAB method only appeared at the high molecular mass position on the electrophoresis bands. However, the signals that were induced by genomic DNA extracted through SDS method appeared both at the high and low molecular mass position on the electrophoresis bands. Genomic DNA extracted through SDS method was fit for Southern analysis.
