超速冷冻8-细胞鼠胚

用3.5M和2.5M DMSO作冷冻保护剂进行超速冷冻8-细胞鼠胚。用前者作保护剂的鼠胚,无论在体外培养和体內发育的情况都好于后者。前者中的8-细胞鼠胚解冻后的存活率为78.9％;存活胚胎体外发育到囊胚的比率为83.4％,体內附植率为63.9％,胎儿形成率为43.2％。实验证明,超速冷冻保存胚胎是一种省时、省力、省钱的好方法。

ULTRARAPID FREEZING OF 8-CELL MOUSE EMBRYOS

The 8—cell mouse embryos ultrarapidly frozen in 3.5M DMSO had a significantly better chance to develop both in vitro and in vivo than those ultrarapidly frozen in 2.5M DMSO. The survival rate of embryos after thawing, the rate of survived embryos' developing into blastocysts in culture and the implantation and fetus formation rates in vivo were 78.9%, 83.4%, 63.9% and 43.2% respcclivcly.It was shown that ullrarapid freezing for embryo cryoprescrvation was a simple low-cost and rapid method.
