小型西瓜离体培养技术研究

摘    要:试验为组培诱导四倍体育成无籽小西瓜提供基础条件,进行小型西瓜离体培养技术体系研究.结果表明,无菌苗子叶近轴端为小型西瓜组培诱导丛生芽最佳外植体,最适丛生芽诱导培养基为MS+6-BA 2 mg·L-1+NAA 0.1 mg·L-1;丛生芽在培养基1/2MS+6-BA 2.0 mg·L-1上增殖系数高;分化的小型西瓜不定芽在1/2MS+KT 0.2 mg·L-1+IAA 0.01 mg·L-1的培养基上苗伸长好且苗壮,经壮苗培养的小型西瓜幼苗在1/2MS+IBA 0.3 mg·L-1生根效果最好.

Abstract:The technical system of culture in vitro of mini-watermelon was established in this study.This study showed that the cotyledons paraxial end of aseptic seedlings served as the optimum explants from clustered buds of mini-watermelon induced by tissue culture.The optimal clustered buds-induced medium contained MS+ 6-BA 2 mg·L-1+NAA 0.1 mg·L-1;the proliferation rate of clustered buds was the highest in the medium of 1/2MS+ 6-BA 2.0 mg·L-1;the adventitious buds of mini-watermelon differentiation formed the strong seedlings in the medium of 1/2MS + KT 0.2 mg·L-1 + IAA 0.01 mg·L-1,and the effect on rootedness for seedlings of mini-watermelon was the best in the medium of 1/2MS+IBA 0.3 mg·L-1 by strengthening the seedlings.
