苜蓿子叶节离体培养体系的研究

摘    要:以黑龙江省主栽优质紫花苜蓿为试验材料,建立快速有效的苜蓿子叶节离体再生体系,为便于开展苜蓿遗传体系研究,比较了不同激素组合、苗龄、切割方式以及不同蔗糖浓度对苜蓿再生体系的影响。结果表明,在MS+1.5 mg·L-1 6-BA+3%蔗糖、pH 5.8的条件下启动培养为最佳芽启动培养方式,再转入最佳芽诱导培养基MS+1.0 mg·L-1 6-BA+3%蔗糖、pH 5.8的条件下,苗龄8 d的子叶节出芽率最高,保留两片子叶各切除1/3为最佳切割处理方式,确定生根培养基为:1/2MS+1.0 mg·L-1 IBA+1.5%蔗糖,pH 5.8,0.8%琼脂,整个过程从种子萌发到驯化移栽,仅需8周。

Abstract:The cotyledonary nodes of Medicago sativa in Heilongjiang Province were used as materials to establish a quick and efficient regenerating system of Medicago sativa and study genetic system.The effects of combination of different hormones,seedling ages,splitting ways and sucrose concentration on establishing re-generating system were investigated.The results showed that MS media containing 6-BA of 1.5 mg·L-1,3% sucrose and pH 5.8 was the best,bud shoot media,MS media containing 6-BA of 1.0 mg·L-1,3% sucrose and pH 5.8 was induced media.Seedling age was 8 d,the budding rate was the highest.The cotyledonaries were splitted part of 1/3 separately was the best way.IBA of 1.0 mg·L-1,1.5% sucrose,and pH 5.8 was the most suitable for the shoot regeneration.From the seed germination to transplantation,it needed 8 weeks.
