卷丹与亚洲百合种间杂交花粉管生长的荧光观察

摘    要:利用荧光显微镜对卷丹与亚洲百合品种"多安娜"(Pollyanna)、"精粹"(Elite)、"布鲁拉诺"(Brunello)种间正反交共6个组合授粉后花粉萌发及花粉管生长过程进行观察。结果表明,卷丹作母本的杂交组合授粉后4 h花粉均已萌发,8~24 h花粉管快速生长,48~72 h花粉管到达子房。卷丹作父本的杂交组合授粉后24~96 h内观察到花粉管横向生长、回折缠绕、末端膨大等异常现象,花粉管生长过程中花粉管、花柱道细胞、胚珠中的胚囊出现胼胝质,120 h内很少有花粉管穿过整个花柱到达子房。不同杂交组合花粉萌发及花粉管生长荧光观察结果与田间调查杂交结实率结果有很高的一致性。

Abstract:Pollen tube growth after pollination from six interspecific cross combinations between Lilium lancifolium Thunb.and Asiatic lily cultivars Pollyanna,Elite and Brunello were observed under the fluorescence microscope.It could be found that the pollens started to germinate after pollination 4 h in three combinations when L.lancifolium was used as the female parent.The pollen tubes grew fast after pollination 8-24 h and arrived at the ovary after pollination 48-72 h.When L.lancifolium was used as the male parent,some abnormal phenomena of pollen tubes were observed such as transverse,reflexing or twining growth and swelling on the terminal after pollination 24-96 h.The calloses appeared in the pollen tubes,style cells and embryo sac,and only few pollen tubes reached ovary after pollination 120 h.The results of fluorescence observation of pollen tube growth from different combinations were consistent with the fruit setting rate investigated in the open field.
