| 摘 要： | 通过检测整合酶确定多重耐药大肠杆菌是否含有整合子。试验采用梯度PCR方法确定扩增整合酶最佳温度,以该温度扩增247株菌整合酶,并对PCR产物鉴定测序,确定整合酶类型。结果表明,247株菌中有158株菌扩增出了Ⅰ型整合酶,13株菌扩增出了Ⅱ型整合酶,总共有171株菌扩增出了整合酶基因。表明可通过PCR扩增整合酶的方法监测多重耐药大肠杆菌携带整合子的情况。 |

| Abstract: | To determine whether there were integrons in multidrug-resistant E.coli by detecting integrase,suitable anneal temperature of integrase was determined by gradient PCR method.Integrase was amplified from 247 strains of E.coli at this temperature,then the integrase of PCR-generated DNA fragment was sequenced and its type was determined.The result showed there were 158 strains of E.coli contained Ⅰintegrase and 13 strains of E.coli contained Ⅱintegrase,altogether there were 171 strains of E.coli contained integrase.Therefore,PCR amplifying integrase could monitor integron in multi-drug resistant E.coli. |
