摘    要:根据已知的叶霉病抗性基因Cf-5基因序列,设计了3对嵌套引物。以4份抗病材料和4份非抗病材料DNA为模板,扩增番茄Cf-5基因并比较了抗感材料Cf-5基因的序列,在580、2 879 bp位点分别有T与C、G与T的碱基替换,设计了等位基因特异引物,获得了稳定的等位基因PCR产物,成功地开发了抗叶霉病基因Cf-5的SNP标记,为培育抗叶霉病番茄品种奠定了基础。

Abstract:Based on leaf mould resistant gene Cf-5 sequence,three nested primer pairs were designed for amplifying Cf-5 gene from genomic DNA of four resistant and four susceptible tomato accessions.Comparison of DNA sequences from resistant and susceptible accessions revealed two base substitutions(T to C and G to T at 580 and 2 879 bp,respectively).The specific primers were designed based on the SNP result and obtained a stable SNP marker for Cf-5 gene screening.It is a potential Cf-5 gene marker in tomato breeding.
