雷竹花粉萌发率及贮藏方法研究

摘    要:为更好地开展竹类植物的杂交育种工作,对雷竹花粉的萌发率及贮藏方法进行了研究。结果表明,利用离体花粉萌发法测定的雷竹花粉萌发率远低于氯化三苯四氮唑(TTC)染色法测定的花粉生活力;添加50 mg.L-1硼酸有利于雷竹花粉萌发;不同发育阶段的花粉萌发率有明显差异,以花药完全露出未开裂的花粉萌发率为最高;花粉采集时间以15:30优于9:30;雷竹花粉不耐贮藏,4℃湿藏有利于保持花粉生活力。

Abstract:Pollen germination and storage of Phyllostachys violascens were studied for bamboo crossbreeding.The results were summarized as follows: the pollen germination rate measured by in vitro pollen germination was lower than the pollen viability rate detected by triphenyltetrazolium chloride triphenyltetrazolium chloride(TTC) staining technique;the pollen germination ability could be improved by supplementing with 50 mg · L-1 boric acid in the medium;the pollen germination rate of the anthers completely outcrop but not dehiscent was higher than those of other development stages;as for the pollen collection time,15:30 was better than 9:30;pollen viability of Ph.violascens would be lost quickly during storage,and certain humidity and low temperature at 4℃ could be beneficial to keep the pollen viability.
