罗汉果叶片培养及其不定芽形成的细胞学观察

摘    要:以罗汉果叶片为外植体,探讨了不同基本培养基、不同培养方式、不同植物生长调节剂对不定芽分化的影响,并对不定芽的形成进行了细胞组织学观察。结果表明:基本培养基以改良MS较为适宜,在附加6-BA1.0mg/L+NAA0.5mg/L+2,4-D0.5mg/L的固体培养基中,平均每块愈伤组织再生芽高达5.6个。叶片正面接触培养基的培养方式芽分化率达到90.3%。芽苗在1/2MS+NAA0.2mg/L+IBA0.5mg/L+蔗糖20∥L的固体培养基中诱发生根效果最好。硝酸银对不定芽分化具明显效果,分化率达到86.7%。细胞学观察发现,不定芽起源于叶片紧密愈伤组织表层。

Abstract:The ideal regeneration system of Siraifia grosvenorii(Swingle) C.Jeffery by leaf explants was established.In this study,the effects of different basic media,culture mode and plant growth regulators on shoot regeneration from leaf of Siraitia grosvenirii were investigated,and cytological observation of anatomy of shot regeneration was conducted.The results showed that on the improved medium MS supplemented with 6-BA 1.0 mg/L+/LNAA 0.5 mg+2,4-D 0.5 mg/L,the number of regeneration buds was as high as 5.6 shoots per callus. When the leaf touched the medium directly,shoot regeneration rate reached 90.3%,and plantlets rooting and growth were the best on the solid medium supplemented with 1/2 MS+IBA 0.5 mg/L+(NAA 0.2 mg/L+)20 g/L sugar.Addition of AgNO_3 could stimulate shoot regeneration.The adventitious buds originated from the surface layer of the compact callus.
