双羔型辽宁绒山羊FSHR基因SNP分析的研究

摘    要:分别选择遗传性能稳定的2.5岁的经产双羔、单羔辽宁绒山羊母羊,并以萨能奶山羊为对照。对FSHR基因第10个外显子的1487~1717bp碱基区段进行SNP分析;利用PCR法扩增FSHR第10个外显子的目的片段,用PCR-SSCP法对PCR扩增的目的片段进行单核苷酸多态性分析。结果表明:辽宁绒山羊FSHR基因第10个外显子突变率较大,并且其突变发生在第1606个碱基,由C→T,说明FSHR基因的第10个外显子可以作为双羔性状的标记基因。

Abstract:SNP analysis was conducted from 1 487bp to 1 717bp on the tenth extron of the FSHR gene from biparous and monotonous Liaoning cashmere goat with hereditary stability and of the same age.At the same time, contrastive experiments were carried out on Sanon goat with the same requirements.The genotypeic DNA was abstracted from the blood of the goat chosen in the experiments.The target fragment on the tenth FSH extron was obtained by PCR amplification.Then the polymorphic analysis of mononucleotide was carried out by the way of PCR-SSCP to the target fragment. The results showed that the mutation rate is comparatively high on the tenth extron of goat FSHR gene, and the position of mutation is on the 1606th base, from C to T.This suggests that the 10th extron of FSHR gene can be the labeled gene for the biparous trait.
