碱胁迫下PGPR菌株对水稻萌发及水稻幼苗生长的促生作用——具ACC脱氨酶活性的PGPR在盐碱逆境下的应用初探

摘    要:目的]解决苏打型盐碱稻作区水稻在育秧阶段受盐碱危害而导致的种子发芽势低、发芽不齐、芽尖枯黄、弯曲,幼苗生长不良甚至死亡的难题。方法]用具ACC脱氨酶活性的PGPR菌株处理水稻种子,在不同浓度的碳酸钠(碳酸氢钠)溶液的碱胁迫下进行水稻种子的萌发实验和幼苗的生长试验,测定水稻种子的发芽势、发芽率,水稻幼苗的根长、苗高及不定根数。结果]结果表明:水稻种子用菌株处理后,发芽势和发芽率都显著地提高,在碱浓度为1.25%时水稻种子的发芽势均超过40%,发芽率在80%以上;同时,该处理也有效地拓宽了水稻幼苗在碱胁迫下生存的生态幅,在1.25%的碳酸钠浓度下,水稻种子能萌发并继续生长,而此浓度下未用菌株处理的水稻种子却不能萌发;在各浓度的碱胁迫下,水稻处理组的幼苗根长、苗长和不定根的数目均显著地优于对照组。结论]具ACC脱氨酶活性的PGPR能够有效地解决水稻种子在盐碱胁迫下萌发及其幼苗生长阶段所受到的危害。

Abstract:Objective] In order to solve the difficult problems which resulted from the salt-alkaloid harmfulness in salt-alkali soil rice area,including low rice germination vigor,no neat germination,the withered and curved bud tip,and the bad growth even dead.Method] The seeds were treated by PGPR strains with activity of ACC deaminase to conduct the rice seedling germination experiment and the seedlings growth experiment under the stress of alkali with different concentrations,and the germination vigor,germination rate of rice seeds,the root length,seedling height and lateral root numbers of rice seedlings were also measured.Result] After treating rice with PGPR strains the germination vigor and germination rate significantly improved,the germination vigor of rice seeds was over 40% and the germination rate was more than 80% under alkaloid concentration of 1.25%.The treatment effectively widened the ecological amplitude of rice seedlings survived in the stress of the alkali.The rice seed could germinate and grow at 1.25% Na2CO3,at which the rice seeds without treating with PGPR strains didn't germinate.Under different concentration of alkali,the root length,seedling height and the numbers of lateral root in treating groups of rice were obviously higher than that in CK group.Conclusion] PGPR with activity of ACC could effectively solve the harm of rice seed caused by salt-alkali stress in germination and seedling growth stage.
