摘    要:目的]建立红树莓提取物中鞣花酸含量的HPLC测定方法,为开发鞣花酸保健品和食品提供理论基础。方法]色谱条件:ODSHYPERSLC18柱(100 mm×4.6 mm,5μm),柱温25℃,用CH3OH-三氟乙酸(0.3%)(体积比50∶50)作为流动相,检测波长为254nm,用外标法测定鞣花酸含量。结果]结果表明,鞣花酸含量在0.322~8.050μg范围内与峰面积呈良好的线性关系,r=0.999 9。精密度试验RSD为0.46%,重现性试验RSD为0.97%,稳定性试验RSD为0.99%,平均加样回收率为93.78%,RSD为0.98%,最后测得鞣花酸含量可达230 mg/kg。结论]红树莓中鞣花酸的含量均高于所报道的其他水果,可更进一步对其机理进行研究。

Abstract:Objective] The theoretical basis of the development of health products and food containing ellagic acid extracted from red raspberry was provided through the establishment of the HPLC determination method.Method] The external standard method was used for the content-testing of ellagic acid under the chromatography conditions: 18 column of ODSHYPERSL C18(100 mm×4.6 mm at 5 μm),column temperature of 25 ℃,CH3OH-trifluoroacetic acid of 0.3%(volume ratio of 50∶50) as mobile phase and detection wavelength of 254 nm.Results] The results showed that there was good linearity between the peak area and the content of ellagic acid in the range of 0.322-8.050 μg(r = 0.999 9).Precision RSD was 0.46%;the reproducibility RSD was 0.97%;the RSD in the stability-testing was 0.99%;the average recovery of sample was 93.78%(RSD: 0.98%).And finally,the content of ellagic acid measured was up to 230 mg/kg.Conclusion] The content of ellagic acid in red raspberry was higher than other that in fruits reported and its mechanism should be further studied.
