摘    要:目的]探索滑菇PholiotarmmekoSW-01菌株产漆酶的最佳营养条件及其酶学性质。方法]首先选取不同的碳源、氮源、pH、金属离子进行单因素试验,得到最佳培养基成分,进行四因素三水平的响应面分析。在酶学性质试验中,设置不同温度及pH,测定漆酶的最适反应温度、pH及热稳定性、pH稳定性。结果]最适的碳源和氮源分别为葡萄糖和蛋白胨,最适pH为5,Cu2+能显著提高漆酶的产量。响应面分析结果表明,最佳的产漆酶条件为葡萄糖42.50g/L,蛋白胨2.10g/L,pH5.47,cu2+浓度1.67mmol/L,在此条件下漆酶活性为943.27U。在酶学性质试验中,漆酶的最适反应温度为30℃,最适反应pH为4.8。结论]条件优化能显著提高滑菇漆酶产量。稳定性试验表明,漆酶对温度较敏感,对pH反应不敏感,在弱酸环境中能发挥较高的活性。

Abstract:The optimum conditions (carbon resources, nitrogen resources, pH and metal ions) for the laccase production and the interaction of the four variables (glucose, peptone, pH and copper ion) were processed using one-single-factor experiment and response surface methodolo- gy, respectively. Different temperatures and pH levels were proceed to evaluate the properties of laccase. Result] The optimum carbon re- source was glucose, the optimun nitrogen resource was peptone, optimum pH acid was 5 and the suitable metal ion was copper ion. Optimum conditions for laccase production were as follows: glucose 42.50 g/L, peptone 2.10 g/L, pH 5.47 and copper ion 1.67 mmol/L, and the maximum laccase activity reached 943.27 U. The laccase showed an optimal reaction temperature of 30 ℃ and pH of 4.8. Conclusion] Un- der the optimum conditions, the laccase production was significantly increased. And the stability experimertts showed that laccase was sensitive with temperature and non-sensitive with pH, and had a good laccase activity in acid conditions.
